The invention discloses an interlocking protection method and system of an SPKS, electronic equipment and a readable storage medium. The interlocking protection method of the SPKS comprises the steps that S1, an interlocking system receives an establishment command of a route; s2, the interlocking system judges whether the SPKS protection area associated with the access is in a non-activated state or not; s3, when the SPKS protection area associated with the route is in a non-activated state, the interlocking system judges whether a turnout exists in the route range or not; if no turnout exists in the route range, entering S4; s4, the interlocking system checks whether other conditions for route establishment are met or not; and S5, when other conditions of route establishment are met, the interlocking system establishes a route. The interlocking protection method for the SPKS has the advantages that when the route is established, the turnout state in the route and the state of the SPKS protection area associated with the route are fully applied to interlocking control logic, risks caused by invalid turnout actions due to the fact that route establishing operation is triggered too early can be fundamentally avoided, and safety and economical efficiency are higher.
