PROBLEM TO BE SOLVED: To provide a drive device of a two-wheeled vehicle, which allows sure positioning and can be easily assembled, and a method of assembling the drive device of the two-wheeled vehicle.SOLUTION: A drive device 69 of an electric two-wheeled vehicle 1 comprises: a motor 3 which is provided in an electric motor case 87, has an output shaft 88 protruding outside the electric motor case 87 and generates driving force of a drive wheel; a reduction gear case 85 combined with the electric motor case 87; and a planetary gear mechanism 86 which is provided in the reduction gear case 85 and transmitting driving force from the output shaft 88 to the motor 3 while reducing the same in speed. The output shaft 88 integrally has a sun gear 102 to be meshed with planetary gears 101 of the planetary gear mechanism 86. The reduction gear case 85 has a positioning part 112 which is arranged concentrically with the output shaft 88 and is combined with the electric motor case 87. The dimension L1 of a meshing width between the sun gear 102 and the planetary gears 101 is larger than the insertion depth dimension L2 of the positioning part 112.SELECTED DRAWING: Figure 6
